CD4 gene silencing in HL-60 cells
HL-60 cells (ATCC) were nucleofected using the Cell Line Nucleofector™ Kit V and 0.14 µg - 1.0µg siRNA targeted to CD4. Cells were analyzed 48 hours post nucleofection by flow cytometry and stained for CD4 expression using a PE anti-human CD4 antibody [BD Bioscience Pharmingen, Cat No. 555347]. Cells were gated according to forward/side scatter and propidium iodide staining.
Controls: nucleofection of siRNA against vimentin (color), nucleofection without siRNA oligos (color) and addition of CD4 siRNA oligos without nucleofection (color) shown as an overlay of histograms.
